Response to domestication and selection for growth in the European sea bass (Dicentrarchus labrax) in separate and mixed tanks ArchiMer

Selective breeding of European sea bass (Dicentrarchus labrax) receives a growing interest, as the estimated heritability of growth is medium to high. In this study, we compared the offspring of four groups of sea bass sires, mated with the same wild dams: wild (W), first generation of domestication (D), first generation of mass selection for length (M), first generation of PROSPER-like selection for length (P). The comparison was done both in replicated tanks (separate rearing) and in mixed tanks (mixed rearing) where sire origins were recovered by genotyping of eight microsatellite markers. High levels of mortalities (20%) associated with abnormal swimming have been observed in farmed juveniles of seabass, Dicentrarchus labrax, in Mediterranean coast, southern France. Impacted marine environments lead to metal accumulation in edible marine fish, ultimately impairing human health. Nevertheless, metal accumulation is highly variable among marine fish species. Sea bass Dicentrarchus labrax larvae were hatched and reared for 34 days in the laboratory to test a recent biochemical index (DNA/C) of larval fish nutritional condition. Variations in DNA/C ratio values were monitored during the whole larval ontogenesis of fed individuals and the effect of starvation was assessed at three developmental stages. Three main periods in the relative DNA content variations were determined in close connection with growth patterns during larval ontogenesis.

The ClC-3 chloride channel and osmoregulation in the European Sea Bass, Dicentrarchus labrax ArchiMer

Dicentrarchus labrax migrates between sea (SW), brackish and fresh water (FW) where chloride concentrations and requirements for chloride handling change: in FW, fish absorb chloride and restrict renal losses; in SW, they excrete chloride. In this study, the expression and localization of ClC-3 and Na+/K+-ATPase (NKA) were studied in fish adapted to SW, or exposed to FW from 10 min to 30 days. In gills, NKA-alpha 1 subunit expression transiently increased from 10 min and reached a stabilized intermediate expression level after 24 h in FW. ClC-3 co-localized with NKA in the basolateral membrane of mitochondria-rich cells (MRCs) at all conditions. It is now more an aquaculture than a fisheries species (>96% of the production in 2016), although modern rearing techniques date back only from the late 1980s. The Na+/K+/2Cl(-) cotransporter in the sea bass Dicentrarchus labrax during ontogeny: involvement in osmoregulation ArchiMer

This study combines a cellular and molecular analysis of the Na+/K+/2Cl(-) cotransporter (NKCC) to determine the osmoregulatory role of this protein in different tissues during the ontogeny of the sea bass. We have characterized the complete sequence of the NKCC1 isoform isolated from the sea bass gills and have identified, by immunofluorescence, NKCC1, and other isoforms, within the epithelium of the major osmoregulatory organs. In aquaculture, it is extremely important to determine the composition of fish farm waste to evaluate its potential impacts and to improve its reuse. Near-infra red spectroscopy (NIRS), an alternative to standard chemical analytical techniques, is a quick non-invasive method to assess physical and chemical composition, reducing the cost of routine analysis. Clonal lines are a powerful scientific tool for improved genetic characterization of organisms used in research. Inbred fish lines can be produced in only two generations using uniparental reproduction techniques. Androgenesis, achieved with variable success in several freshwater species, has been attempted in the European sea bass (Dicentrarchus labrax L), a marine fish of commercial and scientific interest. The low yields of progenies inheriting only the paternal genome after UV-irradiation of eggs led to considerations on the occurrence of UV screening compounds in pelagic eggs. Symptoms recalling a nutritional pathology and more precisely a process of lipid peroxydation were observed during the last years in seabass Dicentrarchus labrax and other fishes cultured in tropical marine condition, Ocyurus chrysurus and Lutjanus analis . They included dark coloration, skin ulceration, lethargy, anorexy, emaciation. Histological examinations showed hepatic fatty degenerative lesions, pancreatitis, muscular degeneration and retinal atrophy. Malformations in farmed fish are a prime research topic as it has a major impact on fish welfare and on the economical value of the product. In this study, the genetic component of spine deformities was estimated in European sea bass, a major species of Mediterranean aquaculture, with the use of a posteriori reconstruction of pedigree with microsatellites.
